The export step of the TLM-Tracker workflow allows to save the data achieved for the single cells to a csv- or xls-file. Additionally, growth- and product formation of single cells can be inspected and the according rates can be saved to file.

1 Export

The data export is controlled by the Export panel.

In Screen 2 the growth and fluorescence (product formation) of single cells can be inspected. The data to display are selected with the Display panel. The cells can be selected by a left mouse-click in Screen 1.

per Image

This button opens a dialogue to choose where to save the file. Supported formats are, depending on the operating system, the xls and csv format. The exported data contains following information for every cell in every image:

  • index of the cell [-]
  • index of the parent cell [-]
  • time of the image [h]
  • x- and y-position of the cell [μm]
  • length and width of the cell [μm]
  • area of the cell [μm²]
  • mean fluorescence value [AU]
  • maximal fluorescence value [AU]
  • standard deviation of the fluorescence values [AU]
  • age of the cell [cell divisions]
  • userdata [-]

If a cell is found in multiple images, there will be an entry for every image containing the properties of the cell in this image, but always with the same index for the cell.

Calculated rates

By choosing one the following options the growth- and product formation rates of every cells are determines and exported 'per Image' or 'per Cell'. A more detailes description how these rates are calculated can be found here.

per Image

This button opens a dialogue to choose where to save the file. Supported formats are, depending on the operating system, the xls and csv format. The exported data contains following information for every cell in every image:

  • index of the cell [-]
  • index of the parent cell [-]
  • time of the image [h]
  • growthrate of the cell [h − 1]
  • product formation rate [h − 1]
  • age of the cell [cell divisions]

If a cell is found in multiple images, there will be an entry for every image containing the properties of the cell in this image, but always with the same index for the cell.

per Cell

This button opens a dialogue to choose where to save the file. Supported formats are, depending on the operating system, the xls and csv format. The exported data contains following information for every cell in the image sequence:

  • index of the cell [-]
  • index of the parent cell [-]
  • time of the image [h]
  • growthrate of the cell [h − 1]
  • product formation rate [h − 1]
  • age of the cell [cell divisions]

The difference to the 'per Image' export is that there will be only one entry for each cell in the resulting file. That means, that all the cell property values are averaged from the point of time when a cell arises by division of a parent cell to the point of time when the cell itself divides into two daughter cells. These average values are then saved to file.

Common steps

To export the data achieved in the previous working steps press the 'Rawdata - per Image'-button and choose a filename. If the internally calculated growth- and product formation rates shall be exported use the 'Calculated rates - per Image/per Cell'-buttons. The 'per Image' button saves the rates once for every cell in every image, the 'per Cell' button saves them just once per cell. A cell is regarded as such starting at the point of time of its creation by division of a parent cell and ending at the point of time when itself divides into two daughter cells.
